The Influence of Thidiazuron on Proliferation of Acer x freemanii In Vitro

摘要: Freeman maple (Acer x freemanii E. Murry), a hybrid of red maple (Acer rubrum L.) and silver maple (Acer saccharinum L.), is among the most popular landscape trees grown in the United States. Two cultivars of freeman maple 'Autumn Blaze' and 'Marmo' were introduced from Oregon State by Shandong Institute of Pomology, China, in 2005. Because of their tolerance to cold, superior autumn leaf color and horticultural desirability in Shandong, China, it is significant to speed up propagation and popularization through in vitro culture. Preliminary studies with various concentration combinations of 6-benzylaminopurine (6-BA) and naphthaleneacetic acid (NAA) showed that it is very difficult to induce proliferation for freeman maple. Annual nodal segments with axillary buds of the two cultivars were cultured on Murashige and Skoog (MS) medium without growth regulators to obtain sterile shoots. The sterile shoots were then placed on MS medium supplemented with various concentrations of TDZ (0.001, 0.002, 0.004, 0.01, 0.02, 0.05, 0.1, 0.2, 0.5 mg/L). The results showed that application of TDZ induced direct shoot proliferation. After 6 weeks of initiation culture a total of 3.3 shoots longer than 1 cm were obtained per explant and 2.67 shoots per explant using concentrations of 0.002 to 0.01 mg/L TDZ on 'Autumn Blaze' compared to 2.67 shoots per explant using 0.01 to 0.02 mg/L TDZ on 'Marmo'. At concentrations higher than 0.1 mg/L, TDZ could stimulate the formation of callus with no shoot proliferation and suppress shoot elongation. These results indicated that TDZ effectively stimulates freeman maple shoot proliferation. However, there are individual differences in sensitivity to TDZ between the two freeman maple cultivars. Low concentration TDZ is critical to successful proliferation. TDZ is the optimal cytokinin-like substance for the proliferation of freeman maple in vitro.
